The Effect of Negative Environment Controlled Rearing System(ECRS) to Prevention of Seoul Virus Horizontal Transmission in the Apodemus Agrarius and Rats

Abstract
Seoul virus is one of the member of the genus Hantavirus and a causative agent of Hemorrhagic fever with renal syndrome(HFRS). Seoul virus has been identified in domestic rats inhabiting city areas as well as laboratory rats in many countries. Large quantities of Seoul virus are secreted in the urine, saliva and feces of infected rats. Many investigators and animal breeders have been affected from animal study with Seoul virus infected rodents in laboratory. Negative-type Environment controlled Rearing System(ECRS) is absolutely necessary to such artificial infection research. This study performed to assess preventive ability of horizontal transmission from Seoul virus infected Apodemus agrarius and rats to noninfected Apodemus agrarius and rats in negative ECRS that manufactured in Korea. Furthermore, we investigated the bio-safety of negative ECRS whether horizontal transmission occurs among researchers and breeders.
In usual ECRS, artificially inoculated Apodemus agrarius and rats have antibody titers against Seoul virus. Lower, same floor left and right groups of inoculated Apodemus agrarius and rats were antibody positive. With indirect fluoresence antibody(IFA) method and RT-PCR for antigen test of Seoul virus in lung and kidney, there were positive in lower, same floor left and right groups of inoculated rats, but none of antigen-positive in inoculated Apodemus agrarius.
In improved ECRS, artificially inoculated Apodemus agrarius and rats have antibody titers against Seoul virus. All groups around inoculated Apodemus agrarius and rats were negative. There were negative antigen test of Seoul virus in lug and kidney using IFA method and RT-PCR.
It was considered that improved ECRS have more biosafe effect than usual ECRS in air-borne viral infection researches. In addition, trends of ECRS development should be improve in circumstance equipments such as regularity and balance of air flow, completion of shelter system, miniaturization, weight reduction and noise diminution of blower, convenience of opening and shutting.
